So I just bought a 2003 Ram 2500 crew cab long bed and the trans was bad right when I drove it off lot a used one was put in and now it have a hell of a vibration on take off but feels like it go's away pretty much after it shifts out of first gear iv checked all 3 u-joints and there fine iv checked the rear dif everything looks good in there  dose anyone have any idea what this could be

Did you pull the driveline to check the u-joints or just check for slop? Sounds like a seizing joint.
